Cell Reports | Peng Bo's Team Achieves Xenogeneic Microglia Replacement/Transplantation Across the Full CNS Scale for the First Time

Microglia are important immune cells in the central nervous system (CNS), and genetic defects in these cells can cause or accelerate CNS pathology. Replacing the endogenous microglia in the CNS with exogenous normal cells can slow down the progression of CNS pathology and thus treat the disease.

However, traditional bone marrow transplantation (tBMT) and microglial transplantation (tMT) methods are not efficient in replacing the endogenous microglia in the CNS. Our research group has developed two distinct cell transplantation strategies that can differentiate exogenous bone marrow cells and blood cells into microglia-like cells and efficiently replace the endogenous microglia throughout the CNS. These methods are termed mrBMT and mrPB, respectively. In addition, we have also developed mrMT, which can replace local microglia in the brain with exogenous microglia.
